0094 Effects of Rocking-induced Sleep Enhancement on Wild Type (WT) and APPxPS1 Knock-in (KI) Alzheimer’s Disease (AD) Mice

چکیده
Abstract Introduction Passive rhythmic movement–rocking – is found to enhance sleep, particularly in infants and young wild type (WT) mice. Rocking can potentially sleep at all ages, with the added benefit of being non-invasive, easy translate for human use, negligible side effects. Furthermore, enhancement tools should be especially valuable patients disturbances, a pervasive feature age-related dementia including Alzheimer’s disease (AD). However, rocking has not been explored widely as means enhancement. Therefore, our study explores impact on architecture both WT APP/PS1 KI mice (a model AD). Methods Male C57BL/6J (WT; n=3) APPxPS1 (AD; n=4) (10-12 months old) were kept under 12h:12h light/dark cycle access food water ad libitum. A reciprocating platform was used laterally rock individually housed 1 Hz, rate previously shown younger mice, nine hours during light period. The experimental protocol consisted baseline day, post-rocking rocked only day hours.EEG/EMG signals recorded baseline, rocking, days. Vigilance states (wake, NREM, REM) scored 4-second epochs from EEG/EMG data using standard criteria. Mean bout duration (MBD) time spent each vigilance state estimated compared AD Results In repeated tests (>6 trials per mouse), we observed significant increase NREM (p< 0.001) decrease REM 0.05) percent versus non-rocking days but Additionally, MBD significantly increased, decreased days, Conclusion Our results suggest that potential tool mouse models; investigations larger sample are progress. Future studies will focus cognitive impairment neuropathology, which linked disruption. Support (if any) National Institute Aging (NIA) Grant No:5R01AG068215-02

Sleep-Waking Discharge of Ventral Tuberomammillary Neurons in Wild-Type and Histidine Decarboxylase Knock-Out Mice
Using extracellular single-unit recordings, we have determined the characteristics of neurons in the ventral tuberomammillary nucleus (VTM) of wild-type (WT) and histidine decarboxylase knock-out (HDC-KO) mice during the sleep-waking cycle. The VTM neurons of HDC-KO mice showed no histamine immunoreactivity, but were immunoreactive for the histaminergic (HA) neuron markers adenosine deaminase a...
